Dosing
Osteoporosis 80 mcg subcutaneously once daily into the periumbilical region of the abdomen; treatment duration not to exceed 2 years during a patient's lifetime.

Contraindications
  • History of systemic hypersensitivity to abaloparatide or any component of the product formulation
  • Hypersensitivity to teriparatide or any of its excipients
Adverse Reactions
Most common (>=2%) Hypercalciuria, dizziness, nausea, headache, palpitations, fatigue, abdominal pain upper, vertigo, injection site redness, injection site edema, injection site pain, tachycardia
Serious Orthostatic hypotension, hypercalcemia, hypercalciuria and urolithiasis
Postmarketing Abdominal distension, abdominal pain, constipation, diarrhea, vomiting, decreased appetite, asthenia, lethargy, malaise, hot flush, insomnia, hypersensitivity and anaphylactic reactions, dyspnea, pruritus, rash, generalized pain, blood pressure increased, muscle spasms, injection site bruising and hemorrhage
Most common (>10%) Arthralgia, pain, nausea
Postmarketing Anaphylactic reactions, angioedema, urticaria, drug hypersensitivity, osteosarcoma, hyperuricemia, acute dyspnea, chest pain, muscle spasms of the leg or back, injection site reactions, oro-facial edema
Pharmacology
Abaloparatide is a PTHrP(1-34) analog that acts as an agonist at the PTH1 receptor (PTH1R), activating the cAMP signaling pathway and stimulating osteoblastic activity to promote new bone formation on trabecular and cortical bone surfaces.
Teriparatide is a recombinant human parathyroid hormone analog (PTH 1-34) that binds PTH cell-surface receptors and stimulates new bone formation by preferential stimulation of osteoblastic over osteoclastic activity on trabecular and cortical bone surfaces.
